Array 2 – Intestinal Antigenic Permeability Screen

The Array 2 Intestinal Antigenic Permeability Screen™ is an important initial
screening test for patients with multiple chronic GI and neurologic symptoms,
suspected food sensitivities, chemical intolerances, or a history of
pathogen-induced autoimmune conditions.

What does this test measure?

Cyrex Array 2 is a blood test checking for antibodies (IgG, IgA, IgM) against
proteins that maintain gut lining integrity and a bacterial endotoxin:

  • Occludin and Zonulin are proteins involved in tight
    junctions between intestinal cells.
  • Actomyosin is part of the cell’s cytoskeleton that, if
    damaged, implies a deeper structural breach.
  • LPS (Lipopolysaccharide) is a component of gram-negative
    bacteria’s cell walls (endotoxin) that normally shouldn’t penetrate the gut
    lining.

If a test finds elevated antibodies to occludin or zonulin, it indicates that
the immune system is recognizing these normally internal proteins. This
suggests that tight junctions in the gut may be breaking down and leaking.
Additionally, high levels of LPS antibodies can indicate that bacterial toxins
are entering the bloodstream due to a leaky gut. Overall, this serves as a way
to validate the “leaky gut” hypothesis, which posits that the gut barrier is
compromised and the immune system is responding to gut contents and junction
proteins.

Who is this test most suitable for?

This is used for patients with autoimmune conditions (to see if leaky gut
might be a trigger), chronic inflammatory issues, or persistent GI problems.
If positive, the patient is often advised to do a gut repair protocol (dietary
changes like gluten removal, supplements like glutamine, etc.). It also
distinguishes whether the issue is mainly tight junction opening
(occludin/zonulin) or transcellular epithelial damage (actomyosin), which can
fine-tune treatment focus.
